Most Yeast Plasmid Transformations Performed In A Single Sitting

Michael Smith of Princeton University’s Rose Lab successfully performed 147 yeast plasmid transformations by hand in a single sitting. Each transformation was conducted in a microcentrifuge tube, and then plated onto -URA glucose media.

- each transformation must be successful, including sterile            controls 
- attempter need not wait for yeast to grow, although each transformation must be plated to prove successful transformation
